💥𝐈𝐒𝐒𝐑 𝐒𝐧𝐢𝐩𝐩𝐞𝐭!🐴 𝐒𝐩𝐞𝐫𝐦 𝐊𝐢𝐧𝐞𝐦𝐚𝐭𝐢𝐜 𝐃𝐢𝐟𝐟𝐞𝐫𝐞𝐧𝐜𝐞𝐬 𝐁𝐞𝐭𝐰𝐞𝐞𝐧 𝐂𝐥𝐨𝐧𝐞𝐝 𝐚𝐧𝐝 𝐍𝐨𝐧-𝐂𝐥𝐨𝐧𝐞𝐝 𝐏𝐨𝐥𝐨 𝐀𝐫𝐠𝐞𝐧𝐭𝐢𝐧𝐨 𝐒𝐭𝐚𝐥𝐥𝐢𝐨𝐧𝐬

Cloned horses produced via somatic cell nuclear transfer do not retain the mitochondrial DNA of the founder animal, as mitochondria are derived from the recipient oocyte. Given the established role of mitochondria in s***m motility & energy production, this first-of-its-kind study compared s***m kinematic profiles between cloned & non-cloned Polo Argentino stallions under commercial field conditions.

Study Design:
• 53 Polo Argentino stallions from 7 embryo transfer centers; ages 4–10 years
• Non-cloned stallions with original mitochondria (n=45) vs. cloned stallions with somatic cell nuclear transfer-replaced mitochondria (n=9)
• Semen collected, filtered, & diluted; computer-assisted s***m analysis performed at 5 minutes, 60 minutes, & 120 minutes of incubation at 37°C
• Eight kinematic parameters measured alongside total & progressive motility & velocity subpopulations

Key Results:
• No significant differences in total or progressive motility between groups at any timepoint
• Non-cloned stallions consistently showed superior average path velocity, curvilinear velocity, & amplitude of lateral head displacement (p

It is with heavy hearts that we announce the passing of our stallion, ES Toronto.

Ten years ago, alongside Sheila Armstrong, we imported Toronto and have been incredibly proud to see the impact he made on North American warmblood breeding. An Elite Stallion with the Hanoverian Verband, Toronto was renowned for his exceptional jumping ability, athleticism, temperament, and outstanding pedigree.

Through his offspring, including licensed sons, State Premium mares, and numerous successful sport horses, Toronto leaves behind a lasting legacy that will continue for generations to come.

We are grateful for the years we shared with him and for the influence he had on our breeding program and the wider warmblood community. He will be deeply missed but never forgotten. ❤️🐴

Cryopreserved semen stored in liquid nitrogen is widely assumed to maintain fertility indefinitely, yet emerging reports suggest possible long-term quality decline. This study examined whether extended storage duration affects conventional s***m quality parameters and fertility-associated messenger RNA abundance in commercially cryopreserved stallion semen.

Study Design:
• 18 stallions (Warmblood, Thoroughbred, Lipizzaner); semen cryopreserved between 1998 and 2024; storage duration 0–26 years
• Post-thaw assessment: concentration, motility, membrane integrity, mitochondrial membrane potential, and chromatin condensation
• Messenger RNA extracted from 12 semen batches; abundance of 10 fertility-associated genes measured by quantitative reverse transcription polymerase chain reaction
• Spearman correlation analysis performed between all endpoints and storage duration

Key Results:
• No significant correlations between storage duration and any conventional s***m quality parameter
• No significant correlations between storage duration and messenger RNA quality metrics
• Four genes showed significant negative correlations with storage duration:
•• Protamine 1 (r=–0.338)
•• Protamine 3 (r=–0.373)
•• Peroxiredoxin 6 (r=–0.336)
•• Cysteine-rich secretory protein 3 (r=–0.507)

Conclusions:
• Conventional s***m quality is not meaningfully affected by liquid nitrogen storage for up to 26 years
• Fertility-associated messenger RNA abundance does decline with extended storage duration for select candidate genes
• Major s***m quality changes occur at cryopreservation processing, not during subsequent storage
• Larger sample sizes are needed to confirm messenger RNA findings
